Blood Collection
Samples are collected under ethical protocols using anticoagulants to prevent clotting and maintain cell integrity.

Dilution
Blood is diluted with sterile saline or PBS to optimise density and prepare for separation.

Centrifugation
Diluted blood is layered over Ficoll or Histopaque and centrifuged to isolate the PBMC layer.

PBMC Extraction
The thin white PBMC layer is carefully collected and transferred to a clean tube.

Washing
Cells are washed with PBS or culture medium to remove debris and contaminants.

Counting & Quality Check
Cell number and viability are assessed; only >90% viable PBMCs proceed.

Storage or Use
PBMCs are used fresh or cryopreserved at –196°C for long-term storage and shipping.
